Clomiphene research overview
Clomiphene citrate is a selective estrogen receptor modulator supplied as 30 tablets at 50mg each. It is not a single molecule but a mixture of two geometric isomers, enclomiphene and zuclomiphene, and that composition is central to how it is studied.
The two isomers behave differently. Enclomiphene, the trans isomer, is the predominantly antagonistic component and is the one credited with blocking estrogen negative feedback at the hypothalamus, which raises GnRH pulse frequency and downstream LH and FSH release. Zuclomiphene, the cis isomer, has more agonist character and a considerably longer elimination half-life, so it accumulates across a protocol in a way the trans isomer does not. Research using the mixture is therefore studying a compound whose effective character shifts over time - a fact that makes it a useful subject for isomer pharmacology and a confound for anything else.
