MT-2 research overview
MT-2, or melanotan II, is a cyclic lactam analogue of alpha-melanocyte stimulating hormone. The cyclisation confers resistance to enzymatic degradation that the linear parent hormone lacks, and the analogue binds across the melanocortin receptor family rather than selecting between its members.
That breadth is what makes it a useful laboratory tool and a difficult one. MC1R is associated with melanogenesis and melanocyte research, MC3R and MC4R with central nervous system signaling including appetite and energy balance, and MC5R with exocrine gland function. A compound engaging all of them lets a model survey the family, but attributing an observed effect to one receptor requires either a selective antagonist or a comparison against a more selective agonist. PT-141, a metabolite of MT-2 with a shifted selectivity profile toward MC3R and MC4R, is stocked for that purpose and is the standard comparator in the literature.
