IGF-LR3 research overview

IGF-1 LR3 is an analogue of insulin-like growth factor 1 carrying two changes: an arginine substitution at position 3 and a 13-amino-acid extension at the N-terminus. Both exist to weaken the peptide's affinity for the IGF binding proteins that normally sequester circulating IGF-1.

That matters because in a standard preparation the great majority of IGF-1 is bound to IGFBPs rather than free, and the fraction available to the receptor is a moving target that depends on binding protein concentration in the medium. Reducing binding protein affinity makes the free fraction far more predictable, which is why LR3 appears in receptor signaling work where the concentration reaching the IGF-1 receptor needs to be known rather than estimated. Published models have used it to study cell proliferation and differentiation, PI3K/Akt and MAPK pathway activation, myoblast behaviour and hypertrophy signaling.
